GH safety workshop position paper: a critical appraisal of recombinant human GH therapy in children and adults

چکیده
Recombinant human GH (rhGH) has been in use for 30 years, and over that time its safety and efficacy in children and adults has been subject to considerable scrutiny. In 2001, a statement from the GH Research Society (GRS) concluded that 'for approved indications, GH is safe'; however, the statement highlighted a number of areas for on-going surveillance of long-term safety, including cancer risk, impact on glucose homeostasis, and use of high dose pharmacological rhGH treatment. Over the intervening years, there have been a number of publications addressing the safety of rhGH with regard to mortality, cancer and cardiovascular risk, and the need for long-term surveillance of the increasing number of adults who were treated with rhGH in childhood. Against this backdrop of interest in safety, the European Society of Paediatric Endocrinology (ESPE), the GRS, and the Pediatric Endocrine Society (PES) convened a meeting to reappraise the safety of rhGH. The ouput of the meeting is a concise position statement.

Safety of growth hormone replacement therapy in adults.
Growth hormone (GH) is a powerful metabolic hormone that regulates fuel homeostasis through its protein anabolic and lipolytic actions. The introduction of recombinant human GH has expanded the narrow indication of treating children with severe GH deficiency (GHD) to include a broader target population of children with growth retardation and short stature and adults with hypopituitarism and sev...

Growth hormone (GH) is a polypeptide hormone, secreted by somatotropic cells of the anterior part of the hypophysis. Its application in therapy, first limited to GH deficient children, has now been widened to various other clinical conditions, not necessarily related to short stature. Clinical trials conducted in recent years have proved the safety of its administration in both children and adu...

Recombinant human growth hormone (hGH) was given for 6 months or longer to 68 patients with GH deficiency, 6 with Turner's syndrome and 5 with achondroplasia, during 1986-1994 in a prospective study. The total weekly administered dose of GH was 0.6 u/kg. By random sampling, 29 of 68 GH deficient patients (42.6%) received twice weekly (2 iw) intramuscular (im) injections, 17 (25%) received ...
متن کاملAdvances in Recombinant Human Growth Hormone Replacement Therapy in Adults
Acquired growth hormone (GH) deficiency results from the destruction of normal pituitary and/or hypothalamic tissue, usually from a tumor or secondary to surgical and/or radiation therapy. Diagnostic criteria and clinical sequelae of GH deficiency, although well established in children, are currently areas of active investigation in the adult. It is now apparent that acquired GH deficiency is a...
